William Dafoe Jr. (d. 22 Temmuz 1955, Wisconsin) 2 Akademi Ödülü adaylığı bulunan ABD'li sinema ve tiyatro oyuncusu. Kariyerinde The Last Temptation of Christ (1988), Örümcek Adam film serisi (2002-2007) ve Platoon (1986) gibi kaliteli filmlerde oynamıştır.
2008 yapımı Lars von Trier filmi Antichrist'taki rolüyle Danimarka'nın en önemli ödülü olan Bodil Ödülleri'nde En İyi Erkek Oyuncu ödülünü kazandı. 2014'te yine Lars von Trier'in yönettiği tartışmalı film Nymphomaniac'ta da rol aldı. 2015 yapımı Pasolini filminde de devrinin çok tartışılan yönetmeni Pier Paolo Pasolini 'yi canlandırdı.
80'li yıllarda yönetmen Elizabeth LeCompte ile birliktelik yaşayan Dafoe'nin 1982 doğumlu Jack adlı bir oğlu vardır. 2005'ten beri İtalyan yönetmen Giada Colagrande ile evlidir.
